He announced the bombshell news just hours after the release of his eighth studio album, For All the Dogs, which was released on Friday evening (AEST).
Several scheduled performances have been postponed following the announcement.
He was due to perform in Denver, New Orleans, Nashville and Ohio.
The Canadian-born rapper told a radio programme on Friday that he was going to “lock the door” on the studio for a while.
“I probably won’t make music for a little bit, I’m gonna be honest,” he said.
“I need to focus on my health, first and foremost – and I’ll talk about that soon enough.
“Nothing crazy, but just like, you know, I want people to be healthy in life, and I’ve been having the craziest problems for years with my stomach.”
The break will be indefinite, with the award-winning rapper saying he didn’t “know what a little bit is”.
“Maybe a year or something. Maybe a little longer,” he said.
The 36-year-old posted a clip from the radio programme to his 143 million followers on Instagram, which was accompanied by the caption: “See ya when I see ya.”
Drake has won five Grammys since the release of his first studio album, Thank Me Later, released in 2010.
He took home the gong for Best Melodic Rap Performance at the prestigious awards this year, for his track Wait For U, which featured Future and Tems.
